If the manager and deputy manager  leave the building is it ok for the senior co ordinator to be left in charge of the setting for a couple of hours? She is level 3 and 4 qualified?

I would have thought yes. She's qualified and presumably the ratios are still intact.

Sorry Mia, missed this one. A person left in charge must be a minimum of a level 3. As rose says, ratios must be in accordance with the qualification (and not of those missing if one was EYP.)
